【摘要】:1.1ThisguidecoverstherecommendedguidelinesforperfomingatestforpresenceoforganiccompoundsthatbindtotheAhReceptorandinducetheCYP1Alocusonthevertebratechromosome.Underappropriatetestconditions,inductionofCYP1Aisevidencethatcellshavebeenexposedtooneormoreofthesexenobioticorganiccoimpounds,includingdioxins,furans,coplanarPCBs,andseveralpolycyclicaromatichydrocarbons(PAHs).DetectionofinductionhasbeenmadesimpleandrapidbythestableintegrationofthefireflyplasmidsuchthatAh-receptorbindingresultsintheproductionofluciferase.Luciferaseproductionisafunctionofboththepotencyofthecompound(s)andtheconcentration.ThistypeofReporterGeneSystem(RGS)hasshownconcentration-responserelationshipsusing2,3,7,8-TCDD,5coplanarPCBs,andseveralpolycyclicaromatichydrocarbons(PAHs)(1,2).Thisguidedescribestestconditionsunderwhichsolventextractsofenvironmentalsamples(water,tissue,soil,orsediments)maybetestedforthepresenceofCYP1A-inducingorganiccompounds.1.2Thetestprocedurespresentedinthisguidehavebeenpublishedpreviously(1-3).Thesereferencesshoudbeconsultedtoobtaindetailsregardingtheconstructionandmaintenanceofthecellline,andtheresponseofthecellstovariousorganicsubstances.1.3Alllaboratoryhealthandsafetyproceduresshouldbefollowed.Thisincludestheuseofglasses,gloves,andotherprotectiveclothing,whenhandlingthereagents.Informationontoxicity,handlingproceduresandwasteproceduresshouldbereviewedpriortouseofallchemicals.1.4Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
